MVVAC: MEASLES VACCINE

MVVAC measles vaccine is a live attenuated virus vaccine, produced on primary SPF chick embryo cells. Each 0.5 ml dose contains not less than 1000 PFU of AIK-C strain measles vaccine virus, and not more than 10 mg of antibiotics Erythromycin and Kanamycin. This is a freeze-dried white opaque product manufactured on modern production lines meeting WHO GMP standards. Before use, the vaccine must be reconstituted with the accompanying diluent.

After nearly 4 years of technology transfer from Kitasato Institute under the JICA project (Japan), MVVAC measles vaccine was licensed by the Ministry of Health at the end of 2009. With a capacity of 7.5 million doses/year, the MVVAC measles vaccine production line of POLYVAC has contributed to meeting domestic demand and moving toward export.

COMPOSITION
Each vial of 10-dose measles vaccine reconstituted with 5.5 ml diluent contains per 0.5 ml dose:

D-Sorbitol 0.72%

Hydrolyzed Gelatin 0.36%
Antibiotics (Erythromycin and Kanamycin) ≤ 10 mg

INDICATIONS
Measles vaccine is indicated for active immunization against measles.
Indicated for children from 9 months of age and older, and individuals without measles antibodies.

CONTRAINDICATIONS
– Hypersensitivity to any vaccine component.
– Pregnant women.
– Patients with acute infections, untreated active tuberculosis, or immunodeficiency (except HIV-infected children).

PRECAUTIONS
– Use with caution in individuals with history of allergy or convulsions.
– Do not vaccinate during febrile illness or ongoing immunosuppressive treatment (drugs, blood transfusion, blood products, etc.). Vaccinate children only after at least 3 days fever-free and minimum 4 weeks after end of immunosuppressive therapy.

ADMINISTRATION
Route: Measles vaccine must be administered subcutaneously. Do not inject intravenously.
Dose: 0.5 ml per dose.

Schedule:
– First dose: at 9 months of age or older (per EPI schedule).
– Second dose: booster at 6 years of age.

Preparation and injection method:
– Prepare all injection equipment sterilized to standard.
– Before reconstitution, clean the vial stopper and surface with alcohol.
– Use a 10 ml syringe to withdraw 5.5 ml diluent, inject into a vaccine vial, shake until completely dissolved. Inspect visually to ensure solution is clear, without particles or turbidity.
– Use a 1 ml syringe to withdraw 0.5 ml per dose.
– Subcutaneous injection must follow national EPI guidelines.
– Prevent contamination; use a new syringe and needle for each individual.

ADVERSE REACTIONS
– No serious adverse reactions have been reported after MVVAC administration.
– Possible local reactions: pain, swelling, redness at injection site (mild, resolving within 1–3 days).
– Possible systemic reactions: fever, rash, cough, coryza in some children, lasting 1–3 days, self-limiting.
– Rare cases of mild diarrhea and vomiting (not conclusively linked to vaccine).
– Very rare cases: convulsions, encephalitis, or thrombocytopenia (≤ 1/1,000,000).

DRUG INTERACTIONS
– Do not administer concurrently with immunosuppressive drugs. If long-term high-dose use, delay vaccination for at least 6 months.
– Blood transfusions or gamma globulin containing measles antibodies may reduce vaccine efficacy.
– Tuberculin reaction may be suppressed for 1 month post-vaccination.
– May be administered concomitantly with mumps and rubella vaccines without affecting immune responses.
– Other live attenuated vaccines (e.g., OPV, smallpox, BCG) may interfere with measles vaccine immune response; wait at least 4 weeks after these vaccines before measles vaccination.

STORAGE
– Freeze-dried measles vaccine must be stored at ≤ 8°C, protected from light.
– Diluent should be stored at 2–8°C.
– Keep out of reach of children.

SHELF LIFE
– Freeze-dried vaccine: 24 months from date of manufacture.
– Reconstituted vaccine: store at 2–8°C and use within 6 hours.

PRESENTATION
– Box of 10 vials, each vial contains 10 doses of freeze-dried measles vaccine.
– Box of 10 diluent vials, each vial contains 6 ml diluent.
